Definition / general
- Sex determining region Y (SRY)-related high mobility groupbox 9
- Transcription factor linked to hedgehog pathways, plays a central role in development and differentiation of multiple cell lineages
- Downstream molecule of beta-catenin
- Master regulator of chondrogenesis
Clinical features
- Poor prognostic factor for colon carcinoma (Am J Clin Pathol 2008;130:897, glioma (Med Oncol 2012;29:3484), hepatocellular carcinoma (Diagn Pathol 2012;7:44), non small cell lung cancer (J Exp Clin Cancer Res 2012;31:18)
- Cervix: SOX9 methylation is frequently involved in cervical carcinogenesis (Oncol Rep 2013;29:125)
- Prostate: acts as critical downstream effector of ERG in TMPRSS2:ERG fusion+ prostatic adenocarcinoma (J Clin Invest 2013;123:1109)
- Soft tissue: expressed in Schwann cells from neurofibromatosis 1 tumors (Hum Pathol 2011;42:434)
- Testis: Sertoli cell maturation requires functional androgen/AR signaling to repress SOX9 and AMH expression; impairment of androgen/AR signaling promotes SOX9 mediated AMH production, leading to impairment of Sertoli cells in Sertoli cell only azoospermic patients (PLoS One 2013;8:e76303)
Uses by pathologists
- Distinguish mesenchymal chondrosarcoma (SOX9+) from small cell osteosarcoma (SOX9-, Hum Pathol 2010;41:653)
